Effect of Withanolide-A on Mice Subjected to Simulated Chronic Jetlag/Shift Work

Objective: Circadian rhythm is maintained by central and peripheral clocks that regulate an organism’s physiological functions. Frequent trans-meridian travel, irregular sleep-wake cycle, light exposure during the rest period, rotating shift work, other factors cause desynchronization of clocks, impairing social occupational behavior. Withanolide-A (WA) used for its powerful nootropic neuroprotective properties.Methods: Effects 13 days consecutive pre-dosing WA (10 μmol/kg/day) on circadian disruption caused due to 9 h advance-delay chronic jetlag light-dark was studied re-entrainment rate analyzed through locomotor activity recording mice using ClockLab setup, learning memory Y-maze Barnes maze tests. In hippocampus, gene expression <i>PER1</i> <i>BDNF</i> assessed reverse transcription-quantitative polymerase chain reaction. The effect treatment also superoxide dismutase, catalase, reactive oxygen species (ROS), lipid peroxidation in hippocampal tissue.Results: results showed leads faster better performance genes upregulated treated group compared groups hippocampus suprachiasmatic nuclei. Moreover, WA-treated produced fewer ROS radicals, there less group.Conclusion: These suggest may act as a chronobiotic agent took transient cycles re-entrain after shift. Neuroprotective property supported learning-memory behavioral tests, studies biochemical assays.
